摘要
目的:观察应用cyclophosphamid对兔下颌骨缺损的变化,探讨CTX在兔下颌骨愈合过程中影响。方法:健康的新西兰大白兔24只,体重2.0~2.4kg。在其下颌骨体部制造1.5mm×10mm的部分骨缺损模型,随机分成A、B两组、不限雌雄,每组12只。术后第2天,A组腹腔注射CTX,20mg/kg,B组腹腔注射生理盐水,10mL/只。注射1次/d,共1周。在术后不同时间点行X线片、骨密度、HE染色及免疫组织化学染色观察,并测血清钙素水平。结果:术后4周B组的骨密度较A组高(P〈0.05)。B组术后骨缺损X线评分在1、4、8周明显高于A组(P〈0.05)。术后B组骨缺损部位的新骨生成面积百分比均比A组高(P〈0.05)。B组术后骨缺损处Ⅱ胶原(ColⅡ)蛋白分泌较A组增多(P〈0.05)。术后1、4、8周B组血清骨钙素水平均较A组明显增高(P〈0.05)。结论:在骨缺损的愈合过程中,应用CTX可起延迟骨缺损愈合的作用。
Objective:To observe the changes of rabbit mandibular defect cyclophosphamide and to explore affection of cyclophosphamide in rabbit mandibular defect healing process.Methods:Twinty-four healthy New Zealand white rabbits weighing 2.0-2.4kg were used,which were caused by the mandibular body 1.5x10 mm part bone defect.They were randomly divided into A,B groups,12 in each group(male or female).A group of days between the second postoperative day group A intraperitoneally injected with 20mg/kg CTX,group B was intraperitoneally injected with 10 mL physiological saline.At different time points bone mineral density,X-ray film,immunohistochemical staining,and serum calcium levels were measured.Results:Bone mineral density after 4weeks,bone mineral density of group B was higher than that of group A.X-scores of bone defect after 1w,4w,8wof higher group B than the group A.B group of bone defect after surgery at the percentage of new bone area were higher than A group.B group of bone defect after surgery at the colⅡ protein secretion compared with A group increased.1w,4w and 8wafter the serum osteocalcin levels were significantly higher than A group.Conclusion:During bone defect healing process,applications of CTX can play a role in delayed healing of bone defects.
