摘要
目的:观察虎杖醇提物、水提物对阿尔茨海默病小鼠模型学习记忆能力的影响,并初步探讨其作用机制。方法:采用侧脑室注射β-淀粉样蛋白片段1-42(Aβ1-42)建立AD小鼠模型,通过Morris水迷宫实验检测小鼠学习记忆能力,使用试剂盒检测小鼠脑组织总抗氧化能力、乙酰胆碱酯酶活性、丙二醛和一氧化氮含量。结果:虎杖醇提物、水提物均不同程度地改善模型小鼠的学习记忆能力,提高总抗氧化能力,以醇提物效果更好,醇提物还能抑制乙酰胆碱酯酶活性,降低丙二醛和一氧化氮含量。结论:虎杖能改善Aβ致AD小鼠模型的学习记忆能力,其作用机制可能与抗氧化、减少炎症介质的产生和调节胆碱能系统等多方面综合作用有关。
Objective:To observe the influence of giant knotweed on learning and memory in AD model mice,discuss about improving learning and memory mechanisms.Methods:The mice Alzheimer's Disease model was developed by injecting Aβ1-42into lateral ventricle.By Morris water maze test,observe the effect on learning and memory ability,testtotal antioxidant capacity,the content of MDA,AChE and NO in brain.Results:In treating groups by giant knotweed,the ability of learning and memorizing were improved,total antioxidant capacity was increased,MDA was decreased and the content of Ache and NO were reduced.Conclusion:Giant knotweed has protective effects on AD mice induced by Aβ,its mechanisms are probably owning to antioxidation and inhibiting the activity of AChE and NO.
