摘要
线粒体细胞内氧化应激的平衡会影响线粒体DNA(mt DNA)的生物合成,导致mt DNA突变和水平的改变,线粒体基因突变导致三羧酸循环障碍,引起胰岛β细胞分泌胰岛素不足导致糖尿病。同一个体不同组织mt DNA的水平不同,外周血mt DNA水平的改变与2型糖尿病的关系尚无定论。该文将探讨mt DNA突变的研究进展,mt DNA单倍群涉及糖尿病及其并发症的病理学,以线粒体为靶向位点的药物或者治疗对于预防和治疗2型糖尿病具有巨大潜力。
The balance of oxidative stress in mitochondrial cell affects mitochondrial DNA biosynthesis,which leads to mitochondrial gene mutations and mt DNA content change. Mitochondrial gene mutations lead to the Krebs cycle disorder,which causes the islet beta cells secrete insulin deficiency,and leads to diabetes. Quantities of mt DNA vary between individuals and tissues within an individual. Whether peripheral blood mitochondrial DNA content is a good predictor of type 2 diabetes is still unconfirmed. Here is to discuss the research progress of mitochondrial mutations,and discuss each haplogroup related to diabetes and the pathology of different complications. The drug or treatment targeting mitochondrial has great potential for the prevention and treatment of type 2 diabetes.
