摘要
目的研究线粒体DNA5178A/C多态性在2型糖尿病人与正常人中的分布情况,以及线粒体DNA5178A/C多态性与2型糖尿病人血脂水平的相关性。方法采用聚合酶链反应(PCR)和限制性片段长度多态性(RELP)分析法对2型糖尿组(174例)及正常对照组(207例)进行基因分析,同时测定2型糖尿病组患者的血脂水平。结果线粒体DNA5178A基因型在2型糖尿病组及正常对照组中的比例分别为35%(61/174)、32%(66/207),5178C基因型的比例分别为65%(113/174)、68%(141/207);2型糖尿病组中5178A基因型与5178C基因型血清HDLC、LDLC、TG、apoA、apoB、Lp(a)水平无显著性差异,5178A基因型血清胆固醇水平低于5178C基因型,两型之间有显著性差异(P<0.05)。结论5178A/C多态性与2型糖尿病的发病未见相关性,但5178A基因型在2型糖尿病人中可能有抗动脉粥样硬化作用。
Objectives To study the prevalence of mitochondrial DNA 5178A/C polymorphism in type 2 Diabetes mellitus patients and healthy controls among Chinese Han population of HuBei Province and its association with serum lipid levels in type 2 diabetics. Methods PCR and RELP techniques were used to examine 174 patients with type 2 diabetes(diabetic group) and 207 healthy people (control group) to analyse the mitochondrial gene and detect the serum lipid levels in patients with type 2 diabetes mellitus.Results The frequency of 5178A in diabetic group and control group were 35% and 32% respectively and in the 5178C were 65% and 38% respectively. There were no significance difference between the two groups, for the frequencies of 5178A/C polymorphism genotypes The serum levels of LDL C、HDL C、TG、apoA、apoB、Lp(a) had no significance difference in the 5178A and 5178C but the serum TC level was significantly lower in the 5178A than 5178C.Conclusion the MtDNA 5178A/C polymorphism may not be related to the etiology of type 2 diabetes. However,5178A seems to have an antiatherogenic effect, at least in type 2 diabetic.
