荷梗中的黄酮类化合物及其细胞毒活性 被引量:5

Studies on flavonoids from stems of Nelumbo nucifera Gaertn and their cytotoxic activities

摘要 研究植物莲Nelumbo nucifera Gaertn干燥茎的黄酮类成分及其细胞毒活性,为进一步开发利用荷梗提供依据。采用大孔树脂、硅胶和Sephadex LH-20、C18柱色谱等方法分离纯化,并运用波谱方法对所分离的化合物进行结构鉴定。采用MTT法对所分离得到的化合物进行HL-60癌细胞毒活性实验。从荷梗中分离鉴定了12个黄酮类化合物,分别为鼠李秦素-3-O-β-D-葡萄糖苷(1)、木犀草素-3',4'-二甲基-7-O-β-D-葡萄糖苷(2)、山柰酚-3-O-β-D-吡喃木糖(1→2)-β-D-葡萄糖苷(3)、槲皮素-3,3'-O-二葡萄糖(4)、1,8-二羟基-3,7-二甲氧基口山酮(5)、异鼠李素-3-O-β-D-葡萄糖苷(6)、山柰酚(7)、异鼠李素(8)、槲皮素(9)、紫云英苷(10)、金丝桃苷(11)、1-羟基-3,7,8-三甲氧基口山酮(12)。所有化合物均首次从荷梗中分离得到,化合物1-5为首次从莲科植物中分离得到。在样品溶液浓度为1×10^-5mol·L^-1的条件下,化合物2-4,6对BEL-7402癌细胞体外生长的抑制率分别是67.36%,53.25%,57.78%,60.13%和52.11%,表明上述化合物对人肝癌BEL-7402细胞具有较明显的体外细胞毒活性。 This research is to investigate study the flavonoids from stems of Nelumbo nucifera and the cytotoxic activities of isolated compounds. The constituents were separated by column chromatography,and their structures were elucidated by spectroscopic data analyses. The isolated compounds were evaluated for cytoxic activities by MTT method. Twelve compounds were isolated and identified as rhamnazin-3-O-β-D-glucopyranoside( 1),luteolin-3',4'-dimethylether-7-O-β-D-glucoside( 2),kaempferol-3-O-β-D-xylopyranosyl-( 1→2)-O-β-D-glucopyranoside( 3),quercetin-3,3'-di-O-β-D-glucopyranoside( 4),1,8-dihydroxy-3,7-dimethoxyxanthone( 5),isorhamnetin-3-O-β-D-glucopyranoside( 6),kaempferol( 7),isorhamnetin( 8),quercetin( 9),astragalin( 10),hyperoside( 11) and 1-hydroxy-3,7,8-trimethoxyxanthone( 12). All compounds were isolated from stems of this plant for the first time,and compounds 1-5 were firstly isolated from the family nelumbonaceae. Compounds 2-4 and 6 showed significant cytotoxic activities against BEL-7402 carcinoma cell lines at a concentration of 1 × 10^-5mol·L^-1with the inhibitory rate of 67. 36%,53. 25%,57. 78%,60. 13% and 52. 11%,respectively.
