木瓜多糖对CCl_4所致小鼠急性肝损伤的保护作用 被引量:9

The Protective Effect of Stauntonia Chinensis Polysaccharide on CCl_4-induced Acute Liver Injuries in Mice

摘要 目的:研究木瓜多糖对CCl4所致小鼠急性肝损伤的保护作用。方法:昆明种小鼠50只随机分成空白对照组、病理模型组和木瓜多糖低(50 mg/kg)、中(100 mg/kg)、高(200 mg/kg)三个剂量保护组,灌胃饲养7 d,对照组腹腔注射调和油溶液,其余各组腹腔注射0.15%CCl4调和油溶液,24 h眼球取血分离血清,测血清ALT活性;处死小鼠制备肝匀浆,测定肝匀浆中MDA、GSH、GSH-PX、SOD和NO的含量,同时取肝脏组织进行病理学检查。结果:木瓜多糖各剂量保护组均能降低血清ALT活性,抑制肝组织GSH、GSH-PX、SOD的降低和MDA、NO的升高,减轻肝组织的病理损伤,与模型组比较差异有统计学意义(P<0.01)。结论:木瓜多糖对CCl4所致小鼠急性肝损伤具有明显保护作用。 Objective To investigate the protective effect of stauntonia chinensis polysaccharides( SCP) on CCl4 induced acute liver injuries in mice. Methods Fifty Kunming mice were randomly divided into five groups( n = 10) : control group,pathological model group,and low( 50 mg / kg),medium( 100 mg / kg) and high( 200 mg / kg) dosage of SCP groups. After feeding for 7 days,mixed-edible-oil solution was intraperitoneally injected into the control group,while the other groups were injected with 0. 15% CCl4 mixed with mixed-edible-oil. Sera were collected from mice 24 h later to determine the activity of serum alanine transaminase( ALT). Mice were then sacrificed to prepare liver homogenate. The levels of liver malondialdehyde( MDA),glutathione( GSH),glutathione peroxidase( GSH-Px),superoxide dismutase( SOD) and nitric oxide( NO)were determined. The pathological changes of livers were also examinated. Results Compared with model group,the serum ALT activity was significantly decreased,the decreasing levels of GSH,GSH-Px,SOD and NO were restored and close to normal levels. Moreover,the increasing MDA were obviously inhibited in all SCP groups( P〈0. 01). Meanwhile,the pathological injury of livers were all reduced in three SCP groups( P〈0. 01). Conclusion SCP could protects mice from CCl4-induced acute liver injury.
