摘要
目的:观察香菇多糖对CC l4肝损伤小鼠NO、TNF-α含量的影响,探讨其对CC l4肝损伤的保护作用。方法:50只昆明小鼠分对照组、模型组、香菇多糖高、中、低浓度组。香菇多糖组每天50、100、200 mg/kg香菇多糖灌胃饲养8 d,对照组腹腔注射调和油溶液,其余各组腹腔注射0.15%CC l4调和油溶液,24 h眼球取血分离血清,处死小鼠制备肝匀浆,测血清ALT活性,计算肝体指数,测定肝匀浆中MDA、NOS、NO、TNF-α的含量。结果:香菇多糖组能降低血清ALT活性和肝体指数,降低肝细胞中NOS活性,减少MDA、NO和TNF-α的含量,与模型组比较有统计学意义(P(0.05~P(0.01)。结论:香菇多糖通过加速清除自由基降低MDA的含量,减轻CC l4肝损伤所致炎性反应,降低肝体指数,抑制iNOS活性及TNF-α分泌,降低NO的含量,对CC l4肝损伤起保护用。
Objective To investigate the influence of lentinan on NO and TNF-α contents in mice with CCl4-induced liver injury.Methods Totally 50 mice were randomly divided into five groups:control group,model group,high-,medium-and low-dose of lentinan groups,respectively.Lentinan groups were intragastricly fed lentinan with dose of 50 mg,100 mg,200 mg/kg body weight daily for 8 days.The control group was injected with blend oil and the other groups were intraperitoneally injected with 0.15% CCl4 solution.The serum were prepared from the mice eyes after 24 h.The mice were killed and the livers were homogenated to detect activity of serum ALT.Besides,hepatosomatic index and liver MDA,NOS,NO,TNF-α levels were calculated.Results Lentinan groups could significantly decreased the serum ALT activity,hepatosomatic index,liver NOS activity,NO and TNF-α levels than those of model group.(P<0.05 or P<0.01).Conclusion Lentinan could protect mice from CCl4-induced liver injury.
