摘要
为研究载脂蛋白E受体2(ApoER2)在鹅卵泡发育过程中的表达模式,本研究克隆了鹅ApoER2基因cDNA部分序列,并用Real-time PCR方法检测其在鹅等级前和等级卵泡颗粒层和膜层中的mRNA表达水平。结果表明:克隆得到的鹅ApoER2 cDNA部分序列长848 bp,编码282个氨基酸,预测的鹅ApoER2部分氨基酸序列有4个低密度脂蛋白受体家族的保守结构域,包括低密度脂蛋白受体重复A类结构域、重复B类结构域、YWTD结构域和钙结合表皮生长因子样结构域;ApoER2在卵泡颗粒层和膜层的表达水平均在F1卵泡处显著性升至最高(P<0.05),在其他等级和等级前卵泡颗粒层均趋于平稳(P>0.05),且在等级卵泡中的表达均高于等级前卵泡,在卵泡膜层中的表达先降低后显著性升高(P<0.05)。总体来看,ApoER2在鹅不同等级卵泡颗粒层的表达水平均远高于膜层。以上结果说明,鹅ApoER2蛋白应该具有转运脂质的功能。相比卵泡膜层,ApoER2主要在颗粒层实现其功能,重点表现在等级卵泡上,这可能与鹅卵黄脂质沉积特点有关。
To investigate the expression pattern of apolipoprotein E receptor type 2(ApoER2) of goose, partial ApoER2 cDNA was successfully cloned, and its expression patterns in granulosa and thecal layers of different developmental follicles were researched by real-time PCR. Sequence analyses showed that the partial open reading frame contains 848 bp and encodes for 282 amino acid residues which includes four conserved domains of low density lipoprotein receptor superfamily, such as low density lipoprotein receptor class A domain, LDLa Low density lipoprotein receptor repeat class B, low density lipoprotein receptor YWTD domain and calcium-binding EGF-like domain. Results showed that the expression of ApoER2 was significantly(P〈0.05) to be the highest in granulosa and thecal layers of F1 and became steady(P〉0.05) in granulosa layers of other follicles, however, dominant follicles were higher than prehierarchal follicles in granulosa layers. In the thecal layers, its m RNA abundance tended to decrease gradually and then increase significantly(P〈0.05). In a word, the expression of ApoER2 detected in granulosa layers were higher than that in corresponding follicular thecal layers of different stages of follice development. Results suggested that goose ApoER2 protein may play a role in lipid transport and its functions are displayed mainly in granulosa layers, especially in dominant follicles. Therefore, it may be related to the need of lipid for the formation of yolk in different stages of follicular development.
