瓜子金总皂苷的含量测定及其纯化工艺研究 被引量:3

摘要 目的:建立瓜子金总皂苷含量测定的方法学,研究瓜子金中总皂苷的纯化工艺。方法:采用紫外分光光度法测定瓜子金中总皂苷含量,考察大孔树脂吸附、富集、纯化瓜子金总皂苷工艺。结果:瓜子金总皂苷在测定范围内均显示较好的线性关系(r=0.999 7);精密度、重复性、稳定性试验中RSD值均小于3.00%;加样回收率平均值为99.51%。大孔树脂纯化试验结果表明,70%乙醇洗脱部位总皂苷含量达71.53%。结论:本研究所建立的方法快速、准确,可测定瓜子金总皂苷的含量;大孔树脂可有效富集、纯化总皂苷,为远志属药用植物瓜子金资源的综合利用提供理论支撑。 OBJECTIVE: To establish a method for the content determination and purification technology of total saponins from Polygala japonica. METHODS : The content of total saponins from P japonica was determined by UV visible spectrophotometry. The enrichment and purification of total saponins from P. japonica was determined by macro-porous resin adsorption chromatography. RESULTS: The amount of total sapunins from P japonica showed good linear relationship in the range of determ'mation (r= 0.999 7). RSDs of precision, repeatability and stability tests were all less than 3.00%. The average recovery rate was 99.51%. The content of total saponins in 70% ethanol elution fraction was 71.53% in macro-porous resin purification test. CONCLUSIONS: The established method is rapid, accurate and can be applied for the content determination of total saponins from P. japonica. The enrichment and purification of total saponins can be determined by macro-porous resin, which provide theoretical support for comprehensive utilization of the resource of P. japonica.
