摘要
目的研究槐定碱对慢性心衰大鼠心肌保护作用的机制。方法将60只雄性SD大鼠分为假手术组、心衰组、低(5 mg·kg-1)、中(10 mg·kg-1)、高(20 mg·kg-1)剂量槐定碱干预组(每组12只),运用激光扫描共聚焦显微镜、蛋白印迹等方法研究各组心肌细胞胞浆内咖啡因诱导钙瞬变及钙泵(SERCA2a)表达的变化。结果与假手术组相比较,心衰组大鼠心肌细胞肌浆网内钙瞬变幅度显著下降(18.33±1.52与36.18±2.64,P<0.05);中、高剂量药物干预组大鼠心肌细胞肌浆网内钙瞬变幅度较心衰组明显升高(33.45±2.47、32.19±1.98与18.33±1.52,P<0.05);心衰组大鼠心肌细胞SERCA2a表达较假手术组明显下调,中、高剂量药物干预组大鼠心肌细胞SERCA2a表达较心衰组均明显升高。结论 SERCA2a表达上调促使肌浆网内钙容量增加是槐定碱心肌保护作用的主要机制之一。
Objective study the myocardial protective effect of sophoridine on chronic heart failure in rats. Methods 60 male SD rats were divided into five groups randomly (n =12): sham-operated group,heart failure group,low dose group (5 mg · kg-1 SRI),middle dose group (10 mg · kg -1 SRI/),high dose group (20 mg · kg -1 SRI). The sarcoplasmic reticulum caffeine -induced Ca2+ transients (CCT) were investigated and the expression of Ca2+ -ATPase (SERCA2a) was assessed by using laser scanning confocal microscope and Western - blot. Results The amplitude of Ca2 + transients was reduced in HF group compared with sham group. Caffeine - induced Ca2 + transients (AF/ F0 ) was significantly decreased in HF compared with sham (18.33 ± 1.52 vs 36. 18 ± 2.64,n = 12,P 〈 0. 05). The amplitude of Ca2 + transients was enhanced in middle and high dose group compared with HF group. CCT ( AF/F0 ) was significantly increased in middle and high dose group compared with HF group (33.45 ±2.47,32. 19 ± 1.98 vs 18.33 ± 1.52,n = 12,P 〈0. 05) ,and the expression of SERCA2a was increased in middle and high dose group compared with I-IF group. Conclusion Sophoridine improves heart failure by recovery of sarcoplasmic reticulum calcium capacity,and that is associated with upregulation of SERCA2a.
