摘要
目的在探讨促凋亡因子(Smac)与生存素(Survivin)在胃溃疡组织中的表达。方法收集无癌前病变胃溃疡患者60例、癌前病变胃溃疡患者30例局部病变标本,以实时荧光定量逆转录PCR法检测标本中Smac及Survivin mRNA;免疫组化SP法检测标本中Smac与Survivin表达。结果癌前病变溃疡组织中Smac mRNA表达量为0.74±0.25,低于无癌前病变溃疡组织(1.18±0.14,P<0.05),癌前病变溃疡组织中Survivin mRNA表达量为3.21±1.03,高于无癌前病变溃疡组织(1.73±0.74,P<0.05);凋亡细胞在无癌前病变溃疡治疗前组织中较多,凋亡指数为(18.23±3.62)%,溃疡愈合后凋亡指数为(5.16±0.98)%,治疗溃疡愈合后较治疗前明显降低(P<0.05);在无癌前病变溃疡组织中Smac的阳性表达多为++^+++,在有癌前病变溃疡组织中Survivin阳性表达多为+^++,Smac与Survivin在不同类型的胃溃疡组织中的表达强度等级比较差异具有统计学意义(P<0.05)。结论无癌前病变胃溃疡和癌前病变胃溃疡组织中的Smac与Survivin表达强度不同,无癌前病变胃溃疡组织中Survivin表达强度较高,癌前病变胃溃疡组织中Smac表达强度明显降低。
Objective To investigate expression of pro-apoptotic factor （Smac） and apoptosis inhibitory protein （Survivin） in gastric ulcer.Methods Expression of Smac and Survivin in 60 cases of gastric ulcer without precancerous lesions and 30 cases of gastric ulcer with precancerous lesions were detected by immunohistochemical SP method; expression of Smac and Survivin mRNA were detected by Real time fluorescence quantitative RT-PCR method.Results Expression of Smac mRNA in gastric ulcer patients with precancerous lesions （0.74 ± 0.25） was lower than that in gastric ulcer patients without precancerous lesions （1.18 ± 0.14,P ＜ 0.05）.Expression of Survivin mRNA in gastric ulcer patients with precancerous lesions （3.21 ± 1.03） was higher than that in gastric ulcer patients without precancerous lesions （1.73 ± 0.74,P ＜ 0.05）.Apoptotic index of apoptotic cells after ulcer healing treatment （5.16 ± 0.98） ％ was significantly lower than that of apoptotic cells before ulcer treatment （18.23 ± 3.62） ％ （P ＜ 0.05）.The positive expression of Smac was most ＋＋ ～ ＋＋＋ in gastric ulcer patients without precancerous lesions; positive expression of Survivin was most ＋ ～ ＋＋ in gastric ulcer patients with precancerous lesions,there were significant differences of Smac and Survivin in different types of gastric ulcer tissues intensity （P ＜ 0.05）.Conclusion Expression intensity of Smac and Survivin in gastric ulcer tissues with or without precancerous lesions were different.Expression intensity of Survivin in gastric ulcer tissues without precancerous lesions was higher; expression intensity of Smac in gastric ulcer with precancerous lesions was decreased obviously.
