摘要
研究了四环素 (TC) -Al3+ 配合物与 DNA的相互作用 .吸收光谱和荧光光谱研究表明 ,TC与 DNA无明显的相互作用 ,但在 Al3+离子存在下 ,TC能结合到小牛胸腺 DNA上 .TC先与 Al3+作用 ,生成 TC-Al二元配合物 ,该配合物再与 DNA发生作用 ,形成 (TC-Al) -DNA三元配合物 .TC-Al二元配合物与 DNA的结合常数为 6.5× 1 0 4 L/mol,其配位比 n(TC)∶ n(Al)∶ n(DNA) =1∶ 1∶ 1 .TC-Al配合物与 DNA的键合模式主要是静电相互作用 .溶液的 p H值对 TC-Al二元配合物和 (TC-Al) -DNA三元配合物的形成有一定的影响 .利用 TC-Al配合物与 DNA作用使体系荧光强度增强的性质测定 DNA,其线性范围为 1 .0× 1 0 - 6 ~6.0× 1 0 - 5mol/L,检测限为 5 .0× 1 0 - 7mol/L.分别测定了天然与热变性酵母样品中 DNA的含量 ,结果令人满意 .
The interaction of tetracycline(TC) Al(Ⅲ) complex with DNA was studied. DNA could bind to calf thymus DNA with affinity in the presence of aluminum ions, which have been confirmed by absorption and fluorescence spectra. The increase of peak intensity and red shift of tetracycline in the presence of aluminum ions showed that TC Al binary complex was formed. The spectra changes of TC Al binary complex in the presence of DNA indicate that TC Al binary complex can interact with DNA to form ternary complex with the ratio of n (Al)∶ n (TC)∶ n (DNA)=1∶1∶1. The binding constant of TC Al complex interacting with DNA was 6.5×10 4 L/mol. The formation of binary and ternary complexes could be affected by the pH value of the solution. DNA can also be determined by TC Al binary complex interacting with DNA in the range from 1.0×10 -6 to 6.0×10 5 mol/L with the detection limit of 5.0×10 -7 mol/L.
