摘要
目的 :检测类风湿关节炎 (RA)患者血清中人及猪Ⅱ型胶原抗体水平 ,探讨其在RA发病中的意义。方法 :用自提取的人、猪Ⅱ型胶原包板 ,采用改良的酶联免疫吸附法 (ELISA)测定 1 0 5例RA患者血清抗人、猪Ⅱ型胶原抗体IgG及IgA ,并与正常人 ( 1 4 0例 )及疾病对照组 ( 80例 )比较分析。同时检测患者的类风湿因子(RF)、血沉 (ESR)及C反应蛋白 (CRP)。结果 :RA患者血清中抗人Ⅱ型胶原抗体IgG、IgA阳性率分别为2 5 .3%、2 7.5 % ,抗猪Ⅱ型胶原抗体IgG、IgA阳性率分别为 2 6.9%、2 5 .6% ,显著高于正常人及疾病对照组 (P<0 .0 5 )。两种属Ⅱ型胶原抗体IgG及IgA常同时存在于同一患者中。两种属Ⅱ型胶原抗体与RF、ESR及CRP异常无相关性。结论 :RA患者血清中人及猪Ⅱ型胶原抗体水平升高 ,可能在RA的发病中起着一定的作用 ;但RA患者Ⅱ型胶原抗体阳性与疾病活动指标无明显相关 ,不能作为监测RA活动的指标。
Objective:To detect the autoantibodies of human and porcine typeⅡcollagen and study the pathological roles of these autoantibodies in rheumatoid arthritis(RA).Methods:Using an improved enzyme linked immunosorbend assay(ELISA), anti typeⅡ native collagen antibodies IgG and IgA(include human and porcine) were evaluated in sera from 105 RA patients,140 normal controls and 80 other diseases controls.Results:The IgG and IgA subclass possitive rates of antibodies to native human type Ⅱcollagen were 25.3% and 27.5%, to porcine were 26.9% and 25.6%,respectively.All of these were higher than those in controls.There was no significant difference between the two species. The levels of anti type Ⅱ collagen antibodies had no correlation to RF,ESR and CRP.Conclusion:Dietary collagen could elicit circulating IgG and IgA anti collagen antibodies that cross react with autologous type Ⅱ collagen,but the antibodies have no relationship to the activity of RA.
