摘要
用线粒体(M)专一性活体荧光染色剂罗丹明123显示小鼠受精卵在第一次卵裂周期中M的分布变化。雌原核和雄原核在汇合之前,M在细胞质中呈弥散状随机分布。两原核汇合后,M在核周围略显聚集。第一次卵裂后期,M沿纺锤体微管和2个于核周图集中,但在赤道区域内明显稀少,预示出细胞分裂面的定位。这说明细胞质分裂是收缩环收缩和细胞质结构调整共同作用的结果。2-细胞阶段,M在细胞核周围明显聚集。2-细胞胚受秋水仙素(10μg/ml)作用后,M由环核聚集状态变为在细胞质中弥散分布,而细胞松弛素B没有这种效应。结果表明,在第一次卵裂周期中M的分布形式可能与微管结构和分布有关。
In this study the changes of mitochondrial distribution during the first cleavage cycle of fertilized eggs of mouse was visualized by specific vital fluorescent dye rhodamine l23.Before confluence of male and female pronuclei, mitochondria were randomly scattered in cytoplasm around the pronuclei.After confluence of two pronuclei,mitochondria tend to aggregatad around them.At anaphase of first cleavage,mitochondria were concentrated along the mitotic spindle and around two daughter nuclei but scarcely located in the narrow equatorial region,This pattern of mitochondrial arrangememt implicates that both the contractile ring and the reorganization of equatorial cytoplasm may be responsible for cytokinesis during the first cleavage, In the 2-cell embryos,mitochondria were concentrated in perinuclear region.When they were exposed to colchicine(10 μg/ml)for l h,mitochondria were dispersed throughout the cytoplasm,whereas in the embryos treated with cytochalasin B this case did not occur.These results indicated that the microtubular organization may be involved in the maintenance of mitochondrial distribution during the first cleavage cycle.
