电镜观察结果表明,26℃下茶尺蠖核型多角体病毒形态发生正常,其过程可分成病毒发生基质的形成—核衣壳的形成—病毒粒子的形成—多角体成熟四个阶段,一个细胞核切面可见1~50,甚至更多的多角体,30℃下则其形态发生受阻,开始饲毒后168小时内,宿主中肠上皮细胞和脂肪体细胞核中均未见病毒有关结构,仅观察到染色质凝集块,这应是高温季节该病毒防治效果差的重要原因之一。 此外,在26℃下饲毒所得宿主细胞切片中,还观察到了核内的大量管状物,特长病毒粒子及内含纤维物的具双层膜的囊状物,核内和细胞质中具侧膜的成束纤维状物等特殊结构。
Effect of temperature on the morphogenesis of the nuclear polyhedrosis virus from the tea gcometrid,Ectropis obliqua hypulina Wehrli(EONPV)in the midgut epithelial or fat body cells or the host larvae was studied by electron microscope.The morphogenesis of the NPV was normal as the host larvae fed with the virus and reared at 26℃.It could be divided into four courses as follows:(1)virogenic stromas appeared,(2)nucleocapsids formed,(3)virions formed,and(4)mature polyhedral bodies produced.There was or were from one to fifty or more po-lyhedra in a cut of virus-infected cell nucleus.But the morphogenesis of the virus was inhibited as the host fed with the NPV and reared at 30℃.By 168 hrs.after the beginning of inoculation at 30℃ only chromatin masses were observed within the hosts midgut epithelial or fat body cell nuclei.It was concluded that the inhibition of the morphogenesis of the NPV by high temperature was the main cause for less efficiency of EoNPV in controlling the tea gcometrid during high temperature months.In addation,some special structures,such as tubelike materials,extremely prolonged virion and vacuole with a double envelope containing fibrous materials in nucleus,bundles of fibrous materials with membrane profiles in nucleus or cytoplasm,were observed in the hosts cells infected with the virus at 26℃.
