摘要
绵羊卵巢采自乌鲁木齐市屠宰场 ,卵母细胞体外成熟 2 4~ 2 5h后 ,由体外获能的附睾精子授精 ,5 0h后 ,将卵裂至 2 - 8细胞的绵羊胚胎移入添加了不同浓度细胞因子的培养液中继续培养 ,结果表明 ,加入 5、10、15 μg/ml胰岛素 (Insulin)分别得到 17 46 %、2 3 81%、2 2 95 %的囊胚率 ,与对照组 9 5 2 %相比差异显著 (P <0 0 5 )。高浓度(5 0 0 μg/ml)的粒细胞巨噬细胞集落刺激因子 (Granulocyte -macrophagecolonystimulatingfactor,GM -CSF)可获得2 1 82 %的囊胚率 ,与对照组 7 2 7%相比差异显著 (P <0 0 5 )。胚胎培养液中分别加入 10、10 0、5 0 0ng/ml的白细胞介素 - 3(Interleukin - 3,IL - 3)、白细胞介素 - 8(IL - 8)和神经生长因子 (Nucleargrowthfactor,NGF)与对照组相比 ,囊胚率差异不显著。本研究表明 ,胰岛素、GM -CSF对绵羊体外受精胚胎早期发育有促进作用 ,IL - 3、IL -8、NGF对绵羊胚胎的早期发育无明显作用。
Sheep ovaries were collected from local slaughterhouse,oocytes were selected and cultured for 24~25 h. In vitro capacitating sperms were used for in vitro fertilization.50 hours after in vitro fertilization,2~8 cell embryos were cultured in KSOM to which the five cytokines were added separately.The results show that the addition of insulin at 5,10,15 μg/ml to culture medium obtained 17.46%,23.81%,22.95% blastocyst rates,there were significantly(P<0.05)different compared with that of the control(9.52%).GM-CSF at 500 ng/ml obtained 21.82% blastocyst rates,and it was significantly(P<0.05)different compared with that of the control(7.27%).Development rates to blostocysts of embryos cultured in media supplemented with IL-3,IL-8,NGF(all at concentration of 10,100,500 ng/ml)were no significantly different compared with that of control(P>0.05).It appears that insulin and GM-CSF have the ability to stimulate the development of sheep embryos in vitro (P<0.05).IL-3,IL-8 and NGF at 10,100,500 ng/ml have no effect on development of sheep embryos in vitro (P>0.05).
