固定化酶连续生产半乳糖寡糖 被引量:8

摘要 固定了来源米曲酶的 β -半乳糖苷酶 ,在纤维床反应器中连续生产半乳糖寡糖 在连续反应体系中 ,研究底物浓度、pH、反应温度和停留时间对半乳糖寡糖合成的影响 ,确定了连续生产半乳糖寡糖的最佳工艺条件为底物浓度 4 0 0 g/L ,反应温度 5 0℃ ,pH6 .0 ,停留时间 4 0min 在连续反应 36h时 ,流加 1.5 %D -半乳糖 ,半乳糖寡糖的得率由 39.6 %提高到 6 6 .2 % ,固定化酶反应器能基本稳定地操作 galactosidase from aspergillus oryzae was immobilized for continuous production of galacto-oligosaccharides(GOS) in fibrous bed reactor.The effect of substrate concentration, pH,reaction temperature and retention time on production of GOS was studied.The optimal reaction condition was determined,substrate concentration was 400 g/L;reaction temperature was 50 ℃;pH was 6.0;residience time was 40 min.After 36 h,D-galactose was fed,the product yield increased from 39.6% to 66.2%.The continuous operation was stable for 96 h.
