The significance of HIF expression in neurons of brain diffuse axonal injury

摘要 目的 探讨缺氧诱导因子(HIF)在脑弥漫性轴索损伤中表达的意义。方法利用大鼠头颅瞬间侧向旋转模型造成弥漫性脑损伤,采用免疫组化法测定 HIF-lα的表达,并用TUNEL和免疫组化双重标记法,以及双酶免疫组化法,研究HIF-1α表达与神经细胞凋亡的关系。结果弥漫性轴索损伤后,HIF-lα在丘脑、海马和基底核区神经细胞中呈高表达,并可诱发c-myc和Fas基因表达,启动神经细胞凋亡。 Aim To study the significance of HIF expression in neu- ron of brain diffuse axonal injury( DAT). Methods The brain injury model of 20 adult rats DAI was created by instant head lateral rota- tion. Expression of HIF-lain DAI specimen was detected by immu- nohistochemical ( IHC) staining. The relationship between neuron apoptosis and HIF- 1α expression was investigated by double label- ling technique of TUNEL and IHC staining, and IHC staining with two enzymes. Results The level of HIF-lα expression was high in neurons of thalamus, hippocampus and basal ganglia, accompanied by expression of c-myc and Fas genes which triggered apoptosis of neurons. Conclusion HIF- 1 α may be one of the important factors that induace neuron apoptosis after brain DAI.
