大鼠肾移植急性排异反应中Fas基因的表达与细胞凋亡的关系

摘要 目的:探讨大鼠肾移植急性排异反应中Fas基因的表达与细胞凋亡的关系。方法:建立大鼠同种异体肾移植模型,应用免疫组织化学方法检测Fas基因的表达;应用原位DNA片段未端标记法检测移植肾组织细胞凋亡状态.结果:正常大鼠肾Fas基因主要呈弱阳性或阴性表达,而移植肾Fas基因表达显著增强,且与肾曲小管细胞凋亡指数呈显著的正相关关系(rs=0.512,P<0.001).结论:大鼠移植肾Fas基因的过量表达可引起肾组织细胞的凋亡,导致移植肾结构的破坏和功能的丧失. ve: To investigate the relationship between Fas gene expession and cell apoptosis during acute rejection in rat kidney allograft. Methods: Rat renal allograft model was contracted. Immunohistochemistry was used to detect the expression of Fas gene; In situ labelling apoptotic DNA fragment detection(TUNEL) was used to detect renal allograft tubule cell apoptosis. Results: Fas protein was negative or weakly expressed in normal rat kidney, whereas Fas protein expression was significantly increased in renal allografts during acute rejection. There was a positive relationship between Fas gene exprssion and renal alograft tubule cell apoptosis(rs=0.512, P<0.001). Conclusion: The aberrant expression of Fas gene may led to renal alograft tubule cell apoptosis and destruction of renal structure and loss of function.
