摘要
目的 观察中药半边旗二萜类化合物 5F对体外培养人翼状胬肉成纤维细胞 (humanpterygiumfibroblasts,HPF)增殖的影响。方法 用噻唑蓝比色法 (MTT)观察不同浓度梯度( 4、 8、 1 6、 3 2、 64、 1 2 8μg/ml)的 5F在不同作用时间内 ( 2 4、 48、 72、 96小时 )对HPF增殖的抑制作用。结果 用药 2 4小时浓度 1 6μg/ml开始明显抑制 (P <0 0 5) ;48小时 8μg/ml抑制作用更强 (P <0 0 1 ) ;50 %抑制率 (IC50 )浓度 3 2 μg/ml。同一浓度各时间组抑制率 (IR)与对照组比较 ,在 4μg/ml作用 2 4、 48及 72小时IR与对照组比较差异均无显著性 (P >0 0 5) ,持续作用96小时差异有显著性 (P <0 0 1 ) ;1 6μg/ml以上 2 4～ 96小时各时间组IR与对照组比较差异均有显著性 (P <0 0 1 )。结论 体外培养HPF具较强增殖能力 ,5F对HPE的增殖具明显抑制作用 。
OBJECTIVE To observe the effect of diterpenoid compound 5F of Pteris semipinnata L on the proliferation of cultured human pterygium fibroblasts(HPF ).METHODS Colorimetry MTT was used to detect viability of 5F in different concentration (4, 8, 16, 32, 64 and 128μg/ml) at different time(24, 48, 72, 96 hours ) for inhabitation of proliferation of HPF.RESULTS There was obvious inhibitory effects of 5F on HPFs in the concentration of 16μg/ml for 24h(P<0.05),and was more evident in the concentration of 8μg/ml for 48h (P<0.01) compared with control group. The 50% inhibitory concentration(IC 50) of 5Fwas 32μg/ml. There was no significant differences in the inhibiting rates of HPFs between control group and experimental group at the concentration of 4μg/ml for 24, 28 and 72 hours (P>0.05).There was significant difference at 96 hours(P<0.01).There existed significant differences in the concentration of 16 μg/ml or more from 24 hour to 96 hours compared with control groups(P<0.01). CONCLUSION Cultured HPFs appeared to be obvious proliferative activity.There was a remarkable inhibitory effect of 5F on HPFs, and this effect showed a relativity with time-dependent and dose-dependent.
