半边旗二萜类化合物5F对瘢痕疙瘩成纤维细胞凋亡及Fas蛋白表达的影响 被引量:3

摘要 目的 观察中药半边旗二萜类化合物 5F对瘢痕疙瘩成纤维细胞 (keloidfibrolast,KFB)凋亡及对Fas蛋白表达的影响。方法 体外培养KFB ,①收集以浓度为 32 μg/ml的 5F作用 12、2 4、36、4 8h后的细胞 ;②收集分别用浓度为 8、32、6 4、 12 8μg/ml 5F作用 4 8h后的细胞 ,应用流式细胞仪 (FCM )检测成纤维细胞的凋亡率及Fas蛋白的表达。结果 ① 5F作用 12~ 4 8h成纤维细胞出现明显的凋亡 (P <0 0 1) ,对照组成纤维细胞未见明显凋亡 ;②随着药物浓度的增大Fas蛋白表达也增高 (P <0 0 1)。结论  5F可以诱导KFB细胞凋亡 ,促进Fas蛋白的表达。 Objective To observe the effect of 5F from Pteris semipinnata L on the apoptosis and the expression of Fas in keloid fibroblasts. Methods Keloid fibrolasts were treated with 32 μg/ml 5F for 12, 24, 36 or 48 h and with different concentrations of 5F (8, 32, 64, and 128 μg/ml) for 48 h. The apoptosis and the expression of Fas in keloid fibroblasts were detected by flow cytometry (FCM). Results Obviously apoptotic cells were found when keloid fibroblasts were treated with 32 μg/ml 5F for 12-48 h, but no apoptotic cells were found in the normal control groups. The expression of Fas was enhanced after the fibroblasts were treated with 5F in a concentration-dependent manner (P<0.01). Conclusion 5F could induce the apoptosis of keloid fibroblasts and enhance the expression of Fas.
