摘要
目的 :探讨新型抗癫痫药妥泰是否对新生大鼠缺氧缺血性脑损伤具有保护作用。方法 :将 5 6只 7日龄Wister大鼠随机分为正常对照组 (14只 )、缺氧缺血组 (18只 )、妥泰短期治疗组 (18只 )、妥泰长期治疗组 (6只 )。缺氧缺血后即刻经胃管注入妥泰或生理盐水 ,2 4h后检测各组脑含水量及脑组织钠、钙含量 ,2 8d后观察组织学改变。结果 :缺氧缺血组脑含水量及钠、钙含量明显高于正常对照组 (P <0 .0 1) ;妥泰短期治疗组脑含水量、钠含量明显低于缺氧缺血组 (P <0 .0 1,P <0 .0 5 ) ;长期治疗组组织学检查明显优于缺氧缺血组和短期治疗组。结论 :妥泰能减轻缺氧缺血后脑组织中钠蓄积 ,减轻脑水肿 ,减少神经元死亡 ,具有神经保护作用。
Objective: To investigate whether Topiramate(TPM), a novel antiepileptic drug, is neuroprotective against hypoxic ischemic (HI) brain damage in newborn rats. Methods:Fifty six Wister rats of 7 days old were randomly divided into normal control group(n=14), HI group(n=18) ,short term treated group(n=18) and long term treated group(n=6). TPM or normal saline solution were administered by gastric tube right after HI insult. In each group, brain water content, total natrium (TNa) and total calcium (TCa) were detected in 24 hours and microscopical changes were observed in 28 days. Results: Brain water content, TNa and TCa in HI group were much higher than those in control group (P<0.01). Significant differences were observed between TPM treated group and the control group in terms of brain water content and TNa (P<0.01, P<0.05). Histologic changes in long term treated group were ameliorated compared with those in HI group and short term treated group. Conclusion: TPM may protect nerve cell from HI damage by reducing the accumulation of natrium,alleviating cerbral edema and attenuating neuronal death.
