摘要
目的 :观察体外培养的牙周组织三种主要功能细胞的生物学特性差异。方法 :对人牙龈成纤维细胞 (GFs)、牙周韧带细胞 (PDLCs)及牙槽骨细胞 (ABCs)进行分离、培养、传代后 ,用MTT比色法、碱性磷酸酶测定法、考马斯亮蓝法及茜素红染色法测定其生物学活性。结果 :PDLCs和ABCs在碱性磷酸酶活性 (ALP)及矿化结节形成方面与GFs有显著性差异。结论 :尽管三种细胞具有相同的镜下表现 ,但生物学特性不同 ,则具有不同的表型 ,从而在牙周再生中发挥着不同的作用。
Objective:To observe the different biological characteristics of the three types of cells which comprise the majority of cells in periodontium. Methods: Human gingival fibroblasts(GFs), periodontal ligament cells(PDLCs),alveolar bone cells(ABCs) were isolated,cultured and subcultured. MTT colorimetric assay, alkaline phosphatase activity(ALP)assay, Commasie brilliant blue staining and Dahl McGec Russel's alizarin red stain were used to determine their biological characteristics. Results: Compared with GFs, PDLCs and ABCs showed significant difference in ALP activity and mineralized nodule formation. Conclusion: Though GFs,PDLCs and ABCs had similar morphology under the microscope,they demonstrated different biological behavior in vitro in relation to their different phenotype , and therefore, they may play different roles in periodontal regeneration.
