摘要
利用基因枪法把抗除草剂基因bar基因导入粳稻恢复系超优一号,经PCR,PCR-southern杂交分析,证明外源基因已整合到水稻基因组中,且能稳定遗传;田间喷施除草剂抗性检测结果表明,bar基因在转基因恢复系及后代中均表达,表现抗除草剂,在恢复系与不育系所配制的杂交种中,高效表达,表现高抗除草剂;部分转基因植株T1代分离比符合孟德尔显性单基因的遗传规律,即3∶1规律,部分不符合。
The bar gene transformation of young embryo culture cells and its callus of restoring line of japonica rice through microprojectile has been studied. PCR and PCR-southern analysis indicated that bar gene has been integrated into the rice genome and can transmitted to its later generation T1,T2. It is also noticed that bar gene has expressed not only in the transgene restoring line plants but also in its hybrid plants after spraying of herbicide basta, they all have behaved resistance to herbicide basta, especially in the latter, which are made by male sterile line crossing with transgene restoring line. The ratios of separating of T1 plants of part of transgene rice has been discussed, some are accordance with 3∶1 and others not.
