摘要
采用抑制消减杂交技术构建左侧视网膜剥夺后右视盖基因差异表达的文库 ,研究在无光刺激的条件下视盖发育受阻的分子机制。随机测序 2 1个序列 ,6个序列经反 Northern证实为真阳性 ,3个序列未发现明确的同源序列 ,另外 3个分别与 homosapiens SH3 GL2、G.domesticaus、 transthyretin有较高的同源性。 2 1个序列中 ,11个基因片段被分配序列号。
Using suppression subtractive hybridization technique, the differentially expressed cDNA library for right optic tecta of pigeons was constructed to investigate the molecular mechanism for visual development of the optic tecta without light stimulation. Twenty one of expressed sequenced tags (ESTs) were sequenced randomly. six of EST were proved true positive by reverse Northern Blotting, three of them were found no high homology with genes in Genbank, the rest sharing higher homology with homo sapiens SH3GL2, G.domesticaus, transthyretin. Eleven of ESTs were assigned Accession Number by Genbank. Our data provided useful clues for studying the development of visual system.
