流感病毒B/四川/379/99的核苷酸及氨基酸序列分析

摘要 目的 从分子水平研究四川省乙型流感病毒的变异规律 ,了解其变异特性和变异程度。方法 病毒分离及鉴定方法按照国家标准GB15 994-1995的方法 ;抗原分析用常量半加敏红细胞凝集抑制法 ;核苷酸序列测定采用逆转录双脱氧链终止法 ,DNA序列测定仪读序。结果 发现流感病毒B/四川 /379/99属于B/Yamagata/16 /88毒株系列 ,但与B/北京 /184/93相比抗原性发生了明显的变异 ,核苷酸序列也有 2 7处发生了点突变 ,异致氨基酸序列 12处发生突变 ,其中有 6处位于 70~ 90和 110~ 2 10这两个与抗原决定簇有关的部位。结论 流感病毒B/四川 Objective To study the variant rule of influenza B vincses and to understand its variant characteristic and extent.Methods Isolation of influenza B viruses were performed with chicken embryo cultivation recommended by appendix A in the National Criteria GB15994-1995,its identification with hemaggiutination inhibition test,its antigenicity was analyzed with red cell agglutination inhibition test;nucleotide sequence was determined with didexoy chain termination.Results It was found that influenza B/Sichuan/379/99 viruses belonged to B/Yamagata/16/18 like viruses,but its antigenicity had remarkable variances,as compared with that of B/Beijing/184/93.Moreover,there existed 27 point mutations in its nucleotide sequence and which resulted in 12 point mutations in its amino acid sequence,6 amino acid sequence point mutations existed at 70~90 and 110~210 which related to antigen determinant,Conclusions Influenza B/Sichuan/379/99 is a new mutant strain.
