肺炎克雷伯氏菌SHV28编码基因的克隆、测序及基因定位 被引量:1

摘要 以临床分离的一株肺炎克雷伯氏菌 99 799株为研究对象 ,对其所产生的一种 β 内酰胺酶编码基因的序列、亚型及基因定位进行研究。采用聚合酶链反应 (PCR) ,用 β 内酰胺酶通用引物、blaSHV引物扩增获得 β 内酰胺酶及SHV型酶编码基因的片段 ,将SHV型酶PCR扩增产物克隆入pUCm T载体 ,以双脱氧链终止法测定核苷酸序列 ,再通过GeneBank进行同源性检测确定其亚型和基因所在位置。肺炎克雷伯氏菌 99 799株所含的一种 β 内酰胺酶基因型为SHV型 ,与SHV 2 8编码基因序列完全相同 ,且位于细菌 15 0bp的大质粒上 ,说明重庆地区有SHV 2 8亚型的存在。 To analyse the β lactamase encoding gene in the clinicl resistant isolate Klebsiela pneumoniae 99 799 and identify its subtype. The gene of β lactamase produced by Klebsiela pneumoniae 99 799 strain was amplified by PCR. The purified PCR product was cloned into pUCm T vector and sequenced by dideoxy chain termination composition method. The encoded gene of Klebsiela pneumoniae 99 799 was identified as SHV by PCR. It had the same sequence as the gene encoding SHV 28 and positioned on the 150bp plasmid. Results showed the SHV 28 β lactamase is existing in Chongqing area.
