摘要
目的:表达重组FS-288,制备抗FS抗体,建立特异性的FS双抗体夹心ELISA检测系统。方法:采用表达质粒构建及诱导表达方法制备重组FS-288,杂交瘤技术制备抗FS单抗,酶联免疫方法测定血中FS水平。结果:以抗FS多克隆抗体和单克隆抗体建立了 FS双抗体夹心ELISA法,其敏感度为25 pg/well,正常女性血中 FS水平随月经周期呈周期性变化,在排卵前2 d达峰值,而 acivin A无明显的周期性变化。结论:FS水平随月经周期改变,提示FS可能与性腺轴的内分泌调控有关。
Abstract Objective:To express recombinant follistatin 288(FS-288) and establish a specific two-site enzyme-linked immunosorbent as-say(ELISA) for native follistatin. Methods:The recombinant GST-FS-288 was expressed by using constructed expressive plasmid and inducingby IPTG. The anti-FS monoclonal antibody was prepared using a hybridoma technique. The FS levels in sera were determined by ELISA.Re-sults:The anti-FS monoclonal and polyclonal antibody was prepared by immunizing as antigen using the recombinant FS and purified by usingprotein A affinity chromatography. The sensitivity of the two-site ELISA for native follistatin established by anti-FS monocled and polyclonalantibody was 25 pg/well. Follistatin level in blood of normal women cchanged periodically during the menstrual cycle, its peak was two dsys be-fore ovulation, to contrast activin A level did not markedly changed during the menstrual cycle. Conclusion:The changes of follistatin level inthe serum of normal women during the menstrual cycle suggest the follistatin play an endocrine role in modulating gouadal activities.
