长期深低温保存对骨库异体半关节主要生物学特性的影响 被引量:4

摘要 目的通过对本骨库长期冷冻保存的异体半关节各主要生物学性质的检测,评价骨库储骨的质量。方法取分别储存1、3、5年的异体股骨上半关节,采用三点抗弯曲试验测量其皮质骨最大载荷和抗弯强度;组织形态学和组织化学方法观察其关节软骨、软骨下骨和松质骨形态以及关节软骨基质成分的变化情况;细菌培养的方法检测其有无污染情况。结果各储存年限的皮质骨抗弯强度无显著性差异;骨与软骨组织结构及软骨基质特异性染色无明显改变;细菌培养结果阴性。结论在外包装无破损的情况下,深低温保存的异体骨其主要生物学特性不随储存年限的增加而发生明显改变,其使用是安全有效的。 Objective To ensure the safety and effectiveness of bone allografts, we evaluated the effect of prolonged cryopreservation on half joint bone allografts by investigating the major biological properties.Method Proximal femoral half-joint were obtained from our bone bank after 1,3 and 5 years under -85 ℃ cryopreservation.A three point mechanical bending test was applied on pieces of cortical bone to compare the changing the largest loading and the anti bending strength of fresh and cryoprserved materials. Bacteria culture was exploited to determine the possibility of contamination.Changing of matrix component of articular cartilage and the histology of cartilage, subchondral bone and spongy bone were examined histomorphologically and histochemically.Result All the bacteria culture were negative. There were no significant qualitative histological,or quantitative mechanical differences among the samples.Conclusion If the packages are unabridged , We think that the half joint bone allografts can safely be used after a cryoperservation period of over 5 years and should not be discarded.
