摘要
利用随机扩增多态性 DNA(RAPD)技术分析了 1 4种沙拐枣属 (Calligonum L.)植物 ,通过对 1 6个 Sangon公司十聚体随机引物进行 PCR扩增 ,3个引物能产生多态性带。对 3个引物扩增产生的 45条扩增产物 ,计算单匹配系数 ,应用 UPGMA方法构建亲缘关系树状图。分析结果表明 :(1 )物种间遗传差异明显 ,具有丰富的遗传多样性 ;(2 ) 1 4种沙拐枣属植物明显聚为 4类 。
species of genus Calligonum were analyzed using random amplified polymorphic DNA (RAPD) markers.To assess the relationships among species,16 random decamer oligonucleotide primers from Sangon were used for polymorphic selection.A total of 45 bands amplified from 3 primers were selected for RAPD analyses.The data were used to generate simple matching coefficients and to construct a dendrogram using UPGMA in STATISTICA programs.The results showed that:(1) Distinct genetic differences and extensive genetic diversity were present among the species.(2) 14 species were clustered into 4 groups,this is basically consistent with the results of traditional morphological taxonomy.
