摘要 目的 评价干化学法测定血清中地高辛浓度的可行性。方法 应用美国Johnson&JohnsonClinicalDiagnostics公司生产的Vitros 2 5 0XRC全自动干化学分析仪 ,测定了 2 4例服用地高辛的病人血清 ,利用本方法与雅培公司Tdx/Flex测试仪使用的免疫荧光偏振法进行了比较。结果 地高辛浓度在 0 .5~ 4 .0ng·ml-1范围内有良好的线性关系 (Y =0 .95 6 5X +0 .0 5 6 5 ,r=0 .9989)。日内RSD为 2 .32 %~ 6 .6 4 % ,日间RSD为 2 .5 0 %~ 7.13% ,回收率为 94 .5 5 %~ 10 2 .33%。与免疫荧光偏振法进行比较 ,P >0 .0 5。结论 应用本方法测定血清中地高辛浓度重复性好 ,准确度高 ,与免疫荧光偏振法的测定结果无显著性差异。且需血量少 ,操作简便、快速 。 Aim To evaluate the feasibility of dry chemistry method to measure the concentration of digoxin in serum.Methods The concentrations of digoxin in serum of 24 patients with digoxin therapy were detected simultaneously by dry chemistry method in Vitros 250 XRC (fully automated dry chemistry analyzer made in Johnson & Johnson Clinical Diagnostics Company in American) and fluorescence polarization immunoassay in TDxFLx analyzer (made in Abbott Company).Results The linear range of the dry chemistry method was 0.5~3.9ng·ml -1 (Y=0.956 5X+0.056 5,r=0.998 9,),within and between-run RSD were 2.32%~6.64% and 2.50%~7.13% respectively,Recovery was 94.55%~102.33%.Significant correlation has been found between dry chemistry method and fluorescence polarization immunoassay,and the results measured by two methods have no statistic difference (P>0.05).Conclusion Automatic dry chemistry is a fast,accurate and stable method for digoxin measurement,which is most suitable for routine medical laboratory.
