摘要
目的 建立人前列腺肿瘤细胞系PC 981 0 6 ,为前列腺肿瘤基础研究提供实验模型。方法 由前列腺肿瘤手术标本中取组织块 ,分别包埋在 3只裸鼠右后肢皮下 ,瘤块增殖后再次包埋于另 1只裸鼠皮下 ,取移植瘤进行体外培养。结果 3个移植瘤均增殖 ,裸鼠体内传代后增殖速率加快。体外培养建立的前列腺肿瘤细胞系PC 981 0 6其形态结构、分化程度与原发瘤保持一致 ,染色体形态为人类核型 ,众数维持在 64~ 73之间 ,占 80 % ,细胞周期分析G1 期 53 .2 % ,G2 期1 7.0 % ,S期 2 9.8% ,G2 /G1 =1 .876 ,为亚三倍体细胞 ,细胞群体培增时间为 1 6 .7h ,前列腺特异性抗原 (PSA)表达阴性。结论 PC 981 0 6与原发癌保持相同的生物学特性 ,体外连续传代 1年以上 ,传代 1 2 0代 ,细胞形态不变 ,生长周期恒定 。
Objective To establish a strain of prostate tumor cell line and provide a useful animal model.Methods Prostate tumor specimens were inoculated into right posterior leg s.c of three nude mice.After two passages in the nude mouse,tumor tissue was cultured in vitro.Following the reference,the identification of cell line was fulfilled.Results All of the transplanted tumor grew in the nude mice and the growth was enhanced in the second passage in the nude mice.After the tissue cultured in vitro a stable growth prostate tumor cell line was obtained without change of morphological structure and differentiation.The mode rage of chromosome is 64 73 accounting for 80% and the cycle analysis showed G 1 was 53.2%,G 2 17.0% and S 1 29.8%.The doubling time was 16.7?h.The expression of PSA was negative.Conclusion The prostate tumor cell line PC 98106 was identified to primary cancer cell in biological characters,has been cultured in vitro continually for more than one year and its characters still remained.Therefore PC 98106 may be considered to be a stable cell line.
