摘要
目的 建立人泌尿系肿瘤无限细胞系 ,为泌尿系肿瘤的基础研究提供实验模型 .方法 无菌取下肿瘤手术标本 ,分别采用组织块直接培养法 ,细胞悬液法 ,裸鼠肿瘤移植模型法进行细胞原代培养 .裸鼠皮下肿瘤块发生明显增殖并长到一定程度后 ,再行裸鼠体内传代两次 ,最后取下 F3代组织块进行原代培养 .培养细胞传代超过 2 0代后按建系标准进行检测 .结果 共取 43例标本 ,其中 38例行组织块直接培养 ,18例 F1代培养成功 ,5例 F3代培养成功 ,最长培养时间为 1.3mo;16例行细胞悬液培养 ,10例 F1代培养成功 ,6例 F3代培养成功 ,最长培养时间为 2 .3mo;40例行裸鼠皮下包埋 ,裸鼠体内传代 F1代成功 6例 ,F3代成功 3例 ,该 3例标本行原代培养后建成 3个无限细胞系 :人肾透明细胞癌 RCC- 986 3,人膀胱癌 BC- 6 ,人前列腺癌 PC- 9810 6 .全部细胞传代 1a以上 ,生长稳定 ,传代周期固定 ,其形态结构 ,分化程度与原发瘤保持一致 ,且遗传学特性包括染色体形态仍为人类核型 .结论 与组织块直接培养法和细胞悬液法相比 。
AIM To establish urogenital carcinoma cell lines and provide the tumor models for further research. METHODS Minced tumor pieces directly culture, tumor cell solu-tion culture and nude mice tumor xenotransplantation model were introduced respectively. If the tumor tissues in the nude mouse subcutis appeared to grow and to become very large, it then was passaged in nude mice for another two times. The tumor tissues in the third mouse were cut and cultured in vitro primarily.When the cells were cultured more than 20 passages, they were certified according to the references. RESULTS 43 specimens were abtainted.38 specimens were cut into fragments and cultured in 25 ml glass flasks directly, F1 18 samples,F3 5 samples, the longest survival time was 1.3 months; 16 tumor cell solutions were cultured and F1 10, F2 6, the longest survival time was 2.3 months. 40 tumor tissues were inplanted into nude mice subcutenousely,F1 6, F3 3. The 3 tumor tissues were incised and cultured in vitro. Finaly 3 infinite tumor cell lines were established:Renal carcinoma cell line RCC-9863;bladder cancer cell line BC-6;prostate cancer cell line PC-98106. They were all cultured for more than 1 years and grew steadily. Also they have the human′s morphological structure and heredity characters. CONCLUSION Compared with the tumor fragments and tumor cell solution culture, nude mouse tumor xenotransplantation model is considered to be a better method to establish urogenital infinite carcinoma cell lines.
