摘要
目的 :研究SCP对人乳腺癌细胞生长、抑制效应 ,探讨其抗癌作用机制。方法 :用不同浓度的SCP加入体外培养的人乳腺癌细胞 (MCF - 7)中 ,观察加药后细胞生长和有丝分裂指数 (MI)的变化 ;用MTT法检测其细胞毒作用 ;Hoechst33342 /PI荧光双染色方法检测细胞凋亡。结果 :SCP明显抑制MCF - 7细胞生长 ;IC50值为 1mg/ml;MI于加药后 48h较对照组明显降低 ;凋亡细胞比例在用药后 48h达 63 .3 %。 结论
Objective:To study the effects and its mechanism of shark cartilage preparation on human breast cancer cells line proliferation. Methods: Using experiment in vitro, we added SCP of different concentrations into the medium in which the cells were cultured. The changes of cell proliferation were assessed by MI. Cytoxicity rate was detected by MTT. The number of apoptotic cells was quantitatively assessed by fluorescent light microscope. Results: The cell growth was remarkably inhibited by SCP; the IC50 values of SCP for MCF-7 cells was 1mg/ml;after 48h of exposure to SCP, MI was remarkably reduced.,the rate of Apoptotic cells was increased. Conclusion:Malignant phenotype of MCF-7 was changed and its proliferation ability was remarkably inhibited by SCP. <
