鼻粘膜上皮细胞中缺氧调节血管内皮生长因子及其机制 被引量:2

A study of up-regulates expression of vascular endothelial growth factor in hypoxia of nasal mucosal epithelial cells

摘要 目的:探讨人类鼻粘膜上皮细胞应答组织缺氧,分泌合成血管内皮生长因子(VEGF)的能力,以及缺氧调节VEGF的可能机制。方法:将人类鼻粘膜上皮细胞分别在常氧(21%)和缺氧(1%)条件下进行无血清原代细胞培养3~48小时,另外常氧培养的其中一组加入金属镍共同孵育,采用原位杂交和酶联免疫吸附试验(ELISA)的方法检测各种条件下VEGF的mRNA水平和蛋白质水平。结果:① 缺氧条件下,VEGF的mRNA水平和蛋白质水平明显升高,与常氧组比较P<0.001,且具有时间依赖性;② 加入金属镍共同孵育,VEGF的表达也升高,与常氧组比较P<0.01。结论:在人类鼻粘膜上皮细胞中,缺氧可明显上调VEGF的mRNA和蛋白质表达,它的机制可能是通过含铁血红素氧敏感分子,一个与促红细胞生成素(EPO)调节相似的途径。 Objective: To study the ability of response to tissue hypoxia and induce vascular endothelial growth factor(VEGF) in human nasal mucosal epithelial cells(HNMECs),the regulate mechanism of VEGF in hypoxia condition was investigated. Methods: HNMECs were cultured without serum under normal(21% O2)(group A) or hypoxia(1% O2)(group B) for 3~48% hours,and the third group was normal oxygen+n ickel for culture(group C). The levels of VEGFmRNA and VEGF protein were detected by assay of situ hybridixation and ELISA. Results:(1) The expression of VEGFmRNA and VEGF protein were increased significantly with time-dependence in group B,compare with control group(group A) showed P<0.01.(2) In group C,the expression of VEGFmRNA and VEGF protein were increased than that in group A(P <0.01),it showed that the nickel have ability of stimulate increase of VEGFmRNA and VEGF protein. Conclusions: Under hypoxia,the expressions of VEGFmRNA and VEGF protein appeared up-regulates in HNMECs. Our experimental result showed that the induction of VEGFmRNA and VEGF protein expression by HNMECs in response to hypoxia culture condition should be modulated by an oxygen-sensing hemelike protein similar way to regulation of the erythropietin gene.
