摘要
目的 建立新的抗HCV国家参考品,促进国产抗HCV试剂的发展。方法 从全国各地收集供血员及丙型肝炎病人血样4000多份,应用10多种抗HCV EIA试剂筛选,应用免疫印迹及HCV PCR方法进行确证,并会同26家单位进行会同标定。结果 新参考品由40份抗HCV阴性样品及40份抗HCV阳性样品组成,阳性样品中有单独某一种抗体阳性的样品,有同时含有某两种抗体的样品,也有同时含有4种抗体的样品。结论 新参考品针对试剂存在的问题及发展方向,对研究开发和筛选高质量的丙肝抗原提供了参考数据,并为国产试剂的质量控制提供了标准。
Objective To prepare the new national reference panel for anti - HCV.Methods 4000 sera were collected and detected with more than 10 anti - HCV EIA kits and confirmed using RIBA and HCV UNA PCR, the results were reconfirmed by 26 laboratories.Results The panel was made up with 40 anti - HCV negative and 40 anti - HCV positive samples.Some positive samples contain only single anti - HCV core or NS3 or NS4- antibody, some of them contain 2-4 kinds anti - HCV antibodies. Conclusions The result showed the activity and purity of HCV NS3 antigen used by domestic anti - HCV EIA kits should be improved.
