懒猴高重复顺序DNA的核苷酸顺序分析及其与类α-DNA顺序的比较

摘要 为确知懒猴高重复顺序与灵长类类α顺序间的相互关系,我们将懒猴DNA经EcoRI酶切所得到的高重复顺序DNA的最小片段重组到质粒pUC^(12)上,经转化、筛选、鉴定得到含有懒猴高重复顺序片段的克隆,命名为pLS(EcoR Ⅰ)-1,测定其全部核苷酸顺序为641bp,发现该顺序有与类α顺序的相似性,但变异性较大,我们对此进行了讨论。 In order to determine the relationship between the highly repeated sequence of Lorise and α-sequence of Primates,the smallest fragment LS(EcoR J )-l was inserted into plasmid pUCl2. After transformation, selection and identficatioh, The clone [termed pLg(EcoRI)-1] that contained the highly repeated DNA fragment was obtained. The complete nucleotide sequence of LS(EcoRI)-1 fragment 641 bp in length, was determined by Dideoxynucleotide Termination Sequencing Method with sequenaseTH kit.We analysed the characteristics of the sequence by a microcomputer, and found that there are some similarities and obvious differences with the a sequence of Primates in the LS(EcoRI-l) sequence.The results are discussed.
