摘要
研究了皱褶假丝酵母脂肪酶在AOT/异辛烷反胶团中催化橄榄油水解的性能。发现在25℃,pH7.9,R=[H_2O]/[AOT]=10的条件下,脂肪酶的活力最高。脂肪酶在含水量较低的反胶团中较稳定,当R=5.4,于33℃保存12h,活力可保持89%;底物浓度高达50%(V/V)时仍无抑制;当'水池'内存在40mmol/L的Cu^(2+)、Co^(2+)、Mn^(2+)等金属离子时对酶仅有少许抑制。
The hydrolysis of olive oil catalyzed by Candida rugosa lipase solu-bilized in 'H_2O/AOT/isooctane' reverse micellar solution has been stu-died. The optimal conditions observed for lipase in reverse micelles aret=25℃, pH_(st)=7.9 and R=[H_2O]/[AOT]=10. Lipase in reverse micellesis found to be quite stable. For example, after incubating at 33℃ for12h in AOT/isooctane with R=5.4, the residual activity of lipase re-mains as high as 89%. No inhibition has been found as the substrateconcentration increased up to 50%(v/v), and only a little inhibitionoccurs when 40mmol/L Cu^(2+), Co^(2+) and Mn^(2+) exist in water pools.
