摘要
从各种加工厂采集的污泥、废水样品中,分离出157株菌落呈白色,绒毛状或粉状,皮膜型或脂泥型有脂肪酶活力的菌株。经筛选,获得一株产生高活力脂肪酶的菌株——S863,其发酵液酶活力达400u/ml。对S863菌株进行了形态、培养特征、生理生化特性等鉴定,认为是个新变种,定名为白地霉利用D-阿拉伯糖变种(Geotrichum candidum var.D-arabinosum)。 S863菌株发酵液的离心上清液,经乙醇沉淀,丙酮脱水干燥,得到脂肪酶粗制剂。它在聚乙烯醇橄榄油乳化液系统中,水解橄榄油的最适pH为7.6,最适温度为43℃,在pH 5-9范围内5℃下存放24小时及pH 7.6 40℃保温15分钟,酶活力均未见损失。
A strain of S863 with high lipase activity was isolated from a sampling mud from the oil processing factory. Its lipase activity reached 400u per milliter of culture fluid.
Based on the studies of morphological cuhural, physiological and biochemical characteristics, the strain S863 was assigned as Geotrichum candidum var. D-arabinosum.
The activity keep of the crude enzyme on olive oil was at pH 7.6, 43℃ and was stable in the range of pH 5 to 9 when incubated at 5℃ for 24h, and not inactivited until 40℃ at pH 7.6 for 15 min.
