摘要
目的 比较人结膜上皮细胞接种培养在不同浓度的甘油保存液、不同保存温度及不同保存时间羊膜上的差异。方法 将传代培养的第 2代结膜上皮细胞分别接种在纯甘油及 5 0 %甘油 (DMEM或平衡盐液∶甘油 =1∶1 )保存的羊膜表面 ,观察其体外培养情况。结果 纯甘油保存的羊膜组结膜上皮细胞 48h后即贴壁 ,且生长旺盛 ,而 5 0 %甘油短期保存羊膜组结膜上皮细胞不能贴壁生长。结论 经纯甘油保存的羊膜接种结膜上皮细胞易贴壁生长 ,而采用 5 0 %甘油短期保存 ,则因羊膜上皮尚未脱落 。
Objective To compare of human conjunctiva epithelial cells cultured on amniotic membrane inocubated in different concentration of glycerin preservative fluid, different preservative temperature and different preservative period. Methods The second passage of human conjunctiva epithelial cells were cultured on amniotic membrane inocubated in pure glycerin and 50 percent glycerin (DMEM or balanced salt solution∶glycerin=1∶1) respectively. The cells growth and the condition were observed in vitro. Results In the pure glycerin group, conjunctiva epithelial cells were adhered the amniotic membrane and flourished in 48 hours; in the group of short term period and 50 percent glycerin, conjunctiva epithelial cells fail to adhere the amniotic membrane. Conclusion Compared to the short term period cultured in glycerin mixed with DMEM or balanced salt solution, conjunctiva epithelial cells are more likely to adhere the amniotic membrane cultured in pure glycerin medium, because of the amniotic epithelial cells were fall off.
