摘要
为了观察IL 13对肾小球系膜细胞核因子 κB (NF κB )活性的影响 ,探讨IL 13抑制免疫及炎症反应的分子机制。体外培养的大鼠系膜细胞经LPS激活及不同浓度IL 13处理后 ,利用凝胶电泳迁移率试验 (EMSA )检测系膜细胞NF κB的活性。在含 5 %FCS的RPMI 1640培养状态下的肾小球系膜细胞存在一定的NF κB活性 ;LPS激活后系膜细胞NF κB活性显著增高 ;IL 13可抑制基础状态及LPS激活的系膜细胞NF κB活性。结果表明 ,IL 13可抑制体外培养系膜细胞的NF κB活性。从而抑制NF κB参与调控的细胞因子的表达而最终对系膜细胞炎症效应产生调节作用。
To investigate effects of interleukin 13(IL 13) on the activity of nuclear factor κB(NF κB) in mesangial cells,so as to explore the mechanism of immunosuppressive and anti inflammatory effects of IL 13 in mesangial cells.activity of NF κB in mesangial cells cultured in 5% FCS RPMI 1640 with or without treatment with LPS and/or IL 13 was detected by electropheretic mobility shift assay(EMSA) The activity of NF κB was detected in low level in mesangial cells cultured in 5% FCS RPMI 1640 LPS stimulated the activity of NF κB in mesangial cells IL 13 in 1 ng/ml or 10 ng/ml could inhibit the activation of NF κB induced by LPS in mesangial cells and 10 ng/ml IL 13 could inhibit activity of NF κB in mesangial cells cultured in 5% FCS RPMI 1640 These results suggest that IL 13, as an inhibitory cytokine,may inhibit the expression of inflammatory cytokine through activation of NF κB in mesangial cells
