摘要
目的探讨苦参碱对高糖致大鼠腹膜间皮细胞NF-κB p65表达的影响及其在腹膜炎症发病机制中的作用,为临床防治腹膜硬化提供部分实验依据。方法将30只健康雄性SD大鼠随机分成3组:对照组(n=10,NS组):腹腔每天注射生理盐水30 m l;模型组(n=10,HGC组):腹腔每天注射4.25%的葡萄糖腹透液30 m l+每周1次腹腔注射红霉素6.25万单位;治疗组(n=10,HGM组):腹腔每天注射4.25%葡萄糖腹透液+苦参碱溶液30 m l+每周1次腹腔注射红霉素6.25万单位。8周后分别处死每组各10只大鼠,取腹膜组织以链霉亲和素过氧化物酶(SP)免疫组织化学法行NF-κB p65蛋白检测,并取腹膜组织行HE及M asson染色病检。结果 与NS组比较,HGC和HGM组腹膜间皮细胞NF-κB p65表达上调,但HGC组显著高于HGM组(P<0.05)。结论苦参碱能拮抗高糖引起的NF-κB p65表达上调,减轻腹膜慢性炎症,延缓腹膜纤维化的发生。
Objective To investigate the effect of matrine on high glucose-induced NF-κB p65 overexpression in rat peritoneal mesothelial cells and the possible mechanism in the peritonitis,so as to provide some experimental basis for the prevention and treatment of peritoneal fibrosis.Methods Thirty SD rats were randomly assigned into 3 groups: control group(n=10,NS) established by daily injection of saline 30 mL,model group(n=10,HGC) established through daily 4.25% peritoneal dialysis solution 30 mL and weekly peritoneal injection of erythromycin 62,500 U,and trial group(n=10,HGM) received daily intraperitoneal injection of 4.25% high glucose dialysate added matrine solution 30 mL and weekly erythromycin 62,500 U.Eight weeks later all rats were sacrificed,the expression of NF-κB p65 in the peritoneal tissues was measured by SP method,and the abdominal membrane was stained with HE and Masson method.Results Compared with NS Group,the expression of NF-κB p65 on mesothelial cells was markedly up-regulated in HGC and HGM group,but the expression in HGC group was significantly higher than that in HGM group(P<0.05).Conclusion Matrine might help to resist the overexpression of NF-κB p65 induced by high glucose,alleviate chronic peritoneal inflammatory reaction,and retard the occurrence of peritoneal fibrosis.
