摘要
目的 探讨严重烧伤对小鼠脾脏T淋巴细胞经T细胞抗原受体活化通路的影响及其与伤后T细胞增殖能力和白细胞介素 2 (IL 2 )分泌改变的关系。方法 检测 1 8%Ⅲ度烫伤小鼠T淋巴细胞表面抗原受体TCRα/β、共刺激分子CD2 8阳性表达率 ,及蛋白酪氨酸激酶 (PTK)、蛋白激酶C(PKC)活性改变 ,胞浆游离Ca2 +浓度变化 ,观察伤后不同时相T淋巴细胞增殖功能及IL 2分泌活性 ,并分析上述信号转导分子活性变化与伤后T细胞功能改变的关系。结果 烧伤后T细胞膜受体TCRα/β和共刺激分子CD2 8表达阳性率降低 ,胞膜PTK活性受抑 ,但于伤后 1 6 8h恢复 ,TCRα/β和CD2 8阳性表达率及PTK活性改变与T细胞增殖功能及IL 2分泌呈现显著相关性 ;胞浆游离 [Ca2 +]伤后 1 2h明显降低 ,并持续到伤后 1 6 8h ,胞浆PKC活性出现先升后降的双相改变。结论 烧伤后T淋巴细胞经TCRα/β、CD2
Objective To investigate the changes of murine splenic T lymphocyte activity mediated by T cell antigen receptor (TCR) α/β and CD28 signaling pathway, and explore the proliferation and IL 2 production of T lymphocytes after severe burn injury. Methods After 60 mice were inflicted with 18% TSBA (total body surface area) full thickness scalding by vapor and sacrificed at different time points, the splenic T lymphocytes were collected. The activities of TCR α/β , CD28, membrane PTK, cytoplasmic PKC and calcium concentrations were detected respectively. The proliferation and IL 2 secretion of T lymphocytes were determined. The relationship of the activities of signal transducers mentioned above and the T cell functions was analyzed. Results Compared with control group, membrane TCR α/β , CD28 and PTK activities were suppressed after scalding, but recovered 168 h later, which having a significantly negative correlation with the functions of T lymphocytes. The calcium concentration was lowered significantly 12 h after scalding and remained the reduction to the 168 th hour. Cytoplasmic PKC activity was increased just after the scalding and then decreased 24 h later. Conclusion The suppression of TCR α/β , CD28 and PTK activities may be the main molecular mechanism by which T cells exert dysfunction after scalding.
