光动力作用诱导人鼻咽癌细胞凋亡超微结构形态改变 被引量:8

摘要 目的 探讨光动力疗法 (PDT)对人鼻咽癌细胞株HNE1凋亡的作用。方法 采用从光导纤维输出的氦氖激光 ,功率 15 0mW ,波长 6 32 .8nm ,能量密度 10J/cm2 ,照射含癌光啉光敏剂的鼻咽癌细胞 ,分别在PDT后 3,6 ,12 ,2 4,36和 48h后 ,光镜和电镜观察凋亡细胞的形态学改变 ,并计数细胞凋亡率 (% )。结果 人鼻咽癌细胞株HNE1经PDT 2 4h后出现细胞凋亡的典型形态学改变 ,12h后出现早期凋亡细胞的形态特征 ,光镜和电镜下可见细胞大小不一 ,细胞质浓缩 ,核固缩 ,染色质边集 ,进而细胞裂解和凋亡小体形成。细胞凋亡率分别为 0 % ,1% ,6 % ,71% ,86 %和 95 %。细胞超微结构改变的严重程度依PDT后时间的增加而逐渐递增。结论 PDT对HNE1细胞具有明显杀伤效应 。 Objective To investigate the effects of photodynamic therapy(PDT) on morphological changes of ultrastructure in apoptosis of human nasopharyngeal carcinoma cell line HNE 1.Methods HNE 1 cells were first sensitised by photosensitizer(PSD-007).Then,the He-Ne laser was given,its dose was output power 150 mW,wavelength 632.8 nm and energy density 10 J/cm 2.At the 3rd,6th,12nd,24th,36th,and 48th hour after PDT,the morphological changes of apoptotic cells were observed under the light microscope and electron microscope.The percentage of cell apoptosis was counted.Results The early apoptotic motphological changes in HNE 1 cells appeared after 12 h of PDT.Typical morphological changes were found at 24 h after PDT.By microscope observation,which was an irregular size in cells,cytoplasm condensation,nuclear disintegration,chromatin aggregation in sides and then call disruption,formation of apoptotic bodies.The percentage of apoptotic cell was respectively 0%,1%,6%,71%,86%and 95%.The changes of cell ultrastructure were increased along with the increase of PDT period.Conclusion The study shows that the PDT has an obvious kill effectiveness for HNE 1 cells.It can induce the apoptosis of HNE 1 cells.
