摘要
目的 研究脑转移瘤和胶质瘤中CD4 4v6mRNA和蛋白水平的表达差异 ,探讨CD4 4v6与恶性肿瘤脑转移的相关性。方法 收集脑转移瘤和脑胶质瘤新鲜标本各 3 5例 ,以RT PCR法检测CD4 4v6mRNA的表达 ,同时以WesternBlot法检测其蛋白的表达。以正常脑组织标本作阴性对照。结果 3 5例脑转移癌标本中 ,2 8/ 3 5例 ( 80 % )呈CD4 4v6mRNA的异常高表达 ,2 7/ 3 5例 ( 77.14 % )有CD4 4v6蛋白的过量表达 ,而脑胶质瘤中分别只有 2 / 3 5例( 5 .71% )和 1/ 3 5 ( 2 .86% )有异常表达 ,两组间差异显著。 1例高表达的脑胶质瘤 (GBM)的临床表现为肿瘤多发。结论 CD4 4v6的表达异常在恶性肿瘤脑转移过程中起着重要作用 ,可作为颅内转移性肿瘤临床识别指标。
Purpose To observe the expressions of CD44v6 in metastatic brain tumors and gliomata and to discuss the relationship of CD44v6 and brain metastases. Methods The expressions of CD44v6 mRNA and CD44v6 protein in 35 metastatic brain tumors and 35 gliomata were detected by using reverse transcription polymerase chain reaction and Western blot. Normal brain tissue was studied as negative control. Results The expression of CD44v6 mRNA and CD44v6 protein in metastases were 28/35(80%) and 27/35(77.14%), significantly higher than 2/35(5.71%) and 1/35(2.86%) in gliomata. One glioblastoma (GBM), however, had higher expression. Conclusions CD44v6 is highly expressed in metastatic brain tumors. It may take an important role in metastatic process and provide a useful tool for differentiating primary brain tumors from metastases.
