摘要
目的 建立一种快速、简便 ,适用于基层的恶性疟免疫诊断方法。 方法 将抗恶性疟原虫 HRP- 的单克隆抗体 3A3、2 E7纯化后经配对筛选 ,利用 Dipstick-免疫胶体金技术 ,制成诊断恶性疟的 Dipstick层析条。用该层析条对 115份疟疾患者血样 ,2 0份非疟疾病人血样及 10份正常人血样进行检测 ,评价其敏感性和特异性 ,并对其稳定性、重复性等进行了初步研究。 结果 用该法检测 145份血样 ,其对恶性疟的敏感性和特异性分别为 83.1%及 97.5 % ,已包被抗原、抗体的 Dipstick条在 4℃及室温下可保存 3个月以上 ,对 30份血样重复检测 4次结果完全一致。 结论 Dipstick-免疫胶体金模式快速、简便 ,适用于基层应用 。
Objective To establish a simple, sensitive and reliable diagnostic method for detecting Plasmodium falciparum . Methods Monoclonal antibodies 3A3,2E7 which against HRP Ⅱ antigen secreted by P. falciparum were purified and then applied to dipstick colloidal dye immunoassay. 115 blood samples collected from patients with malaria, 20 from patients from non malaria diseases and 10 normal blood samples were detected by this dipstick method and its sensitivity, specificity, stability and reproducibility were assessed primarily. Results The dipstick diagnostic test was performed easily and results were attained in 8-10 min, the sensitivity and specificity of the test was 83.1% and 97.5% respectively. Dipstick strips coated with antigen and antibody can store at 4℃ and room temperature more than three months, 30 malaria blood samples were detected repeatedly 4 times and results were the same. Conclusion The test is a valuable diagnostic tool for falciparum malaria, especially in field situations requiring rapid diagnosis.
