微血管密度、p53蛋白和PCNA与肝细胞癌的关系 被引量:3

The Relationship of Microvessel Density,p53 and Proliferating Cell Nuclear Antigen to Hepatocellular Carcinoma

摘要 目的 研究肝细胞癌 (HCC)内微血管密度 (MVD )、p5 3蛋白与增殖细胞核抗原 (PCNA )表达之间的关系。方法 采用免疫组化SABC法检测 47例人HCC组织的MVD值、PCNA指数和 p5 3蛋白表达情况。 结果 生存期≥ 5年者的MVD值( 4 1.19± 15 .31)明显低于生存期 <5年者的MVD值 ( 5 4.73± 2 4.72 )。MVD值与患者的年龄、性别、肿瘤的大小、Edemondson分级无关 ,也与PCNA指数和 p5 3蛋白表达无直接相关性 (P >0 .0 5 )。p5 3蛋白在生存期≥ 5年及 <5年的 2组患者中的异常表达率分别为 10 .5 %和 5 0 .0 % ,有非常显著性差异 (P <0 .0 1) ,但其与MVD值无关 (P >0 .0 5 )。低分化HCC的PCNA指数 ( 5 4.14±14 .2 6 )高于高分化HCC的PCNA指数 ( 34.6 2± 6 .16 ) ,有显著性差异 (P <0 .0 5 ) ,但与MVD值无关 (相关系数γ =0 .0 82 ,P >0 .0 5 )。结论 MVD值和 p5 3基因异常表达均与HCC的 5年生存率直接相关 ,但MVD值、PCNA指数和 p5 3蛋白异常表达相互之间无直接相关性。 Objective To study the relationship of microvessel density(MVD) the expression of p53 and proliferating cell nuclear antigen(PCNA) to HCC.Methods Anti-CD_ 34,Anti-p53 and Anti-PCNA monoclonal antibodies were used for immunohistochemical staining on surgical specimens from 47 HCC.MVD was counted and the 5-years survival rate was analyzed retrospectively.Results Blood vessels endothelial cells inside the tumor were strongly stained by anti-CD_ 34 antibody,but not stained in the adjacent liver tissues.The MVD levels of the HCC in patients (41.19±15.31)survivaed for more than 5-years were significantly lower than that(54.73±24.72) in survivors for less than 5-years(P<0.05).Statistical analysis showed that the MVD levels were not correlated with tumor size,Edmondson′s grade,and alpha-fetoprotein level,neither correlated with the expression of PCNA level or p53 protein(P>0.05).However the 5-years survival rate of the p53 positive cases was lower than that of the p53 negative cases(P<0.01),and the difference of PCNA was significant between high and low Edmondson′s grade(P<0.01).Conclusion In HCC,MVD and abnormal p53 expression separately make worse on 5-years survival rate.However,there are no significant correlation among MVD,p53 and PCNA levels each other.
