摘要
根据报道的哺乳动物GH基因序列设计引物 ,首次利用PCR方法扩增和克隆了马鹿GH基因 ,该基因大小约为 1.8kb .对该基因上游 34 0bp核苷酸测序及分析表明 ,测序区域包括 5′侧翼序列 ,第一外显子和部分第一内含子 .第一外显子所编码的所有氨基酸与猪的相比完全相同 .初步认为GH基因在马鹿基因组中为单基因 .
GH gene of Cervus elaphus was amplified by PCR with primers based on the sequences of the reported mammalian GH gene for the first time. The amplified fragment was about 1.8 kb. It was cloned and the partial sequence was sequenced. This sequencing region consists of a 5′-flanking regulatory region, exon I and part of intron I. All amino acids of exon I were the same as those in swine. Comparing the corresponding sequences of GH gene between Cervus elaphus and the reported animals, the authors concluded that the sequencing regions has highly conservation and appears to have only a single growth hormone gene in the genome of Cervus elaphus.
