摘要
以硅胶为吸附剂 ,采用柱层析法分离脑磷脂和卵磷脂 ,对洗脱条件进行了研究 ,使脑磷脂与卵磷脂达到了较好的分离 ,大大减少了洗脱剂用量及洗脱时间 .其最佳洗脱条件为 :洗脱液流速为 40~ 6 0 滴 /m in、梯度差为 1∶ 2 ~ 2∶ 1 的氯仿 /甲醇混合液进行凹形梯度洗脱 .
This paper studies the condition of separating Phosphatidyl Ethanolamine(PE) from Phosphatidyl Cholines(PC) with silica gel absorption column chromatography .By determining the gradient elution process, gradient and flow speed, the PC and PE were separated satisfactorily, the volume of eluent and the time of eluting were reduced greatly. The optimum elution conditions are: flow speed is 40~60 drops/min, gradient is (1∶2)~(2∶1)(methanol /chloroform,V/V) by means of concave gradient elution.
