摘要
以苹果品种“嘎拉”试管苗叶片为试材 ,建立了苹果离体叶片高效直接体细胞胚胎发生体系 ,研究了创伤处理对外植体直接体细胞胚胎发生的效应。结果显示 ,创伤处理提高了外植体直接体细胞胚胎发生率和单位外植体体细胞胚胎产量 ,其中 ,各处理效应大小依次为 :刺伤 >横切 >纵切 ;一定范围内随横切次数和刺点数增加 ,外植体直接体细胞胚胎发生率保持稳定的高水平 ,单位叶片直接体细胞胚胎产量逐渐增加。刺 8点时直接体细胞胚胎发生率由对照的 6 5%增至 1 0 0 % ,直接体细胞胚胎再生数量由对照的 2 9 叶增至 2 6 2
A procedure of efficient direct somatic embryogenesis from in vitro leaves of apple cultivar has been developed. Effect of wounding (pricking,latitudinal cutting and longitudinal cutting) on direct somatic embryogenesis from in vitro leafves of apple was studied.The results showed that effect of pricking was most effective.Regeneration rate and yield of direct embryos increased from 65% to 96% and 2 9/leaf to 26\^2/leaf, respectively when 8 pricks were made on a leaf.
